DNA compaction find may help treat genetic disorders

BHUBANESWAR : In a path-breaking discovery, scientists from Odisha have unearthed some unique physio-chemical properties in ionic liquids (organic salts in liquid state) that can be better alternatives for compaction of DNA to help treat genetic disorders.

After a series of experiments, researchers from National Institute of Science Education and Research (NISER) and Indian Institute of Science Education and Research (IISER) claimed the structure of DNA bends or becomes more spherical as compared to its initial form after molecular interactions with magnetic ionic liquids.

The scientists also found that de-compaction of DNA can be easily achieved by adding NaCl (sodium chloride), also known as salt or common salt, and it has far reaching implications in biomedical applications. The findings on compaction and de-compaction of DNA will help treat genetic disorders such as albinism, Angelman syndrome, ankylosing spondylitis, apert syndrome, congenital adrenal hyperplasia, Huntington’s disease, neurofibromatosis and haemochromatosis besides haemophilia, thalassaemia and several other genetic diseases.

During experiments, we came across some astonishing results involving cholinium-based ionic liquids, which can be better alternatives for DNA compactions,” he said.

An important part of gene regulation in an organism occurs at the level of transcription, which basically works on two principal strategies. But compacted DNA can be useful for inhibition of transcriptional activity. This will be helpful to treat genetic disorders, which are caused in whole or in part by a change in the DNA sequence away from the normal sequence, Prof Biswal told TNIE.

The new phenomenon, the scientists said, can be used as protection against chemical or biochemical stress. It also has promising applications in nano-material templating, stress mitigation, gene regulation, photo-control of gene expression, and safeguarding DNA integrity. Assistant professor of Department of Chemical Sciences at IISER, Berhampur Malay Kumar Rana said the DNA compaction is crucial for cell functioning and involves organising and condensing DNA strands, enabling them to fit within cellular compartments.

The study ‘Compaction and de-compaction of ct-DNA’ has been published in Chemical Science, a peer-reviewed and high-impact journal published by the Royal Society of Chemistry, London. It has been placed on the cover page of the journal.
